ABSTRACT The objective of this study was to compare two alkaloids (antioquine and tetrandrine) with verapamil; knowing that the smooth muscle respond to KCl and relationships with calcium. The effects of antioquine and tetrandrine, was studied in adults Wistar rat with modified methods used in the determination of aorta contractility and compared with verapamil effect in the same assays. The analysis of the effect of a drug or extract on aortic reactivity included maximal relaxation or maximal contraction (Cmax) (Phase 1). In our results, verapamil induced a blockade of 98.7 ± 0.7% (n = 6) in presence of endothelium and 97.9 ± 4.3% in ausence of endothelium, both in phase 1 and in phase 2 of 47.4 ± 4.1% (n = 6) in aortas in the presence of endothelium and... |

Forty mature male rats (Wistar strain) were maintained alive for 40 days, separated in four groups of five rats each (Control, L-NAME, L-NAME + Enalapril, L-NAME + Verapamil). Three aortic wall parameters were analyzed, as follows: the Q A(SMN) (the number of the tunica media smooth muscle nuclear profiles per area), the S V(lamellae) (surface density of the aortic lamellae), and the aortic wall thickness (AWT). Rats of the L-NAME group showed high blood pressure and cardiac hypertrophy. The AWT, the Q A(SMN) and the S V(lamellae) showed the same tendency among the groups. The increased AWT, or the reduced Q A(SMN) and the S V(lamellae) in l-NAME rats were not completely prevented by the administration of enalapril or verapamil. Hypertension causes the AWT... |

Toad envenoming in dogs can cause death by cardiac fibrilation (CVF). Traditional therapy consists mainly of atropine and propranolol, the last one used to prevent the CVF, that is preceded by negative ventricular deflections (NVDs) in the QRS complex of the electrocardiogram. This study intended to verify, comparatively, the lidocaine, propranolol, amiodarone, and verapamil abilities to prevent CVF in experimentally envenomed dogs. Thirty-six dogs were divided into 6 groups (GL, GP, GA, GV, GST, and GSV) with n=6; the dogs were submitted to volatile anaesthesia. The animals of the groups GL, GP, GA, and GV received 0.38g of toad venom through oro-gastric catheter and were treated with the following drugs respectively: lidocaine (4mg/Kg), propranolol... |
